Product Specific Information

Description:
The 11B11 antibody reacts with mouse interleukin-4 (IL-4), a 14 kD cytokine. It functions as a neutralizing antibody. Mouse IL-4 (also known as BCDF, BCGF-1, and BSF-1) is secreted by activated Th2 and NK T cells, and to a lesser extent by Th1 and mast cells. IL-4 promotes B-cell proliferation and differentiation, induces Ig isotype switching, and upregulates MHC class II and CD23 expression.

Reported for use in cytokine neutralization, ELISPOT, and ELISA capture assays.

Applications Tested:
Functional Grade Purified 11B11 antibody has been tested by:

• LAL assay for low endotoxin levels
• ELISA
• Bioassay for neutralization of mouse IL-4 bioactivity

At 0.125 µg/mL, the antibody inhibits 50% of biological effects of 1.0 ng/mL mouse IL-4 in a CTLL-2 cell proliferation assay.

ELISA Use:
Tested as a capture antibody in sandwich ELISA for mouse IL-4, used with biotinylated BVD6-24G2 (Product # 13-7042-85) and recombinant mouse IL-4 (Product # 14-8041-80) as standard.
Recommended concentration: 1–4 µg/mL.
Standard curve: 500 pg/mL – 4 pg/mL.

Target Information

Interleukin-4 (IL-4) is a pleiotropic, immune-modulatory cytokine produced by activated T cells. It acts as a ligand for IL-4 receptor, which also binds IL-13, contributing to overlapping functions. STAT6 mediates IL-4 signaling. The IL-4 gene is located on chromosome 5q near IL-13, forming a cytokine gene cluster (IL-3, IL-5, IL-13, CSF2). IL-4, IL-13, and IL-5 are co-regulated by long-range elements over a 120 kb region. IL-4 delta 2 is a known splice variant.
