Target Information

In the intact cell, DNA closely associates with histones and other nuclear proteins to form chromatin. Chromatin remodeling is critical for transcriptional regulation, and acetylation of nucleosomal histones facilitates this process. Acetylation of lysine residues in histone tails increases DNA accessibility to transcription factors. Several mammalian proteins act as nuclear histone acetylases (GCN5, PCAF, p300/CBP, HAT1, TAF II p250), while histone deacetylases (HDAC1–9) are linked to transcriptional silencing.

HDAC9 and HDAC9a are alternatively spliced isoforms; HDAC9a is 132 amino acids shorter but retains the catalytic domain and deacetylase activity. Both isoforms repress myocyte enhancer-binding factor 2-mediated transcription and are expressed in brain, skeletal muscle, kidney, placenta, and pancreas.
