STAT3는 JAK1과 JAK2에 의해 Tyrosine 705에서 인산화되어 이합체를 형성하고, 핵 내 전위 및 DNA 결합 활성을 나타냅니다. 인간의 STAT3 유전자는 염색체 17q에 위치하며, 여러 전사 인자(c-Jun, CREB 등)와 상호작용합니다.
또한 Rac1 단백질은 STAT3의 활성을 조절하며, PIAS3 단백질은 STAT3의 특이적 억제 인자로 알려져 있습니다. STAT3 유전자의 결실은 생쥐 모델에서 배아 초기 단계의 치명적 결과를 초래합니다.
