Target Information
STAT3는 STAT (Signal Transducers and Activators of Transcription) 단백질 패밀리에 속하며, 수용체 관련 키나아제에 의해 인산화되어 핵으로 이동한 후 전사인자로 작용합니다. 이 단백질은 사이토카인 및 성장인자 자극에 반응하여 다양한 유전자의 발현을 매개하며, 세포 성장과 세포사멸 조절에 중요한 역할을 합니다.
STAT3는 JAK1과 JAK2에 의해 Tyrosine 705에서 인산화되어 이합체를 형성하고, 핵 내 전위 및 DNA 결합 활성을 나타냅니다. 인간의 STAT3 유전자는 염색체 17q에 위치하며, 여러 전사 인자(c-Jun, CREB 등)와 상호작용합니다.
또한 Rac1 단백질은 STAT3의 활성을 조절하며, PIAS3 단백질은 STAT3의 특이적 억제 인자로 알려져 있습니다. STAT3 유전자의 결실은 생쥐 모델에서 배아 초기 단계의 치명적 결과를 초래합니다.
