Target Information
CARMA proteins belong to the membrane-associated guanylate kinase-like (MAGUK) family of proteins that act as molecular scaffolds facilitating assembly of signal transduction complexes.
CARMA1, CARMA2, and CARMA3 share high sequence and functional homology, but their tissue-specific distribution suggests distinct biological functions.
The CARD domain of CARMA3 interacts with BCL10, a positive regulator of apoptosis and NF-κB activation.
CARMA3 is required for EGF-induced NF-κB activation and contributes to tumor growth in vivo, indicating potential as a therapeutic target in EGFR-driven tumors.
