Product Specific Information

This antibody reacts with v-H-ras (within amino acids 120–138) and rodent c-H-ras p21s but not v-K-ras or rodent c-K-ras p21s. It also recognizes human c-H-ras and c-K-ras p21s, but not human c-N-ras.

Preparation:
Rats were injected subcutaneously with Ha-MuSV transformed NRK cells. Sera were screened to identify strong immune responders against p21. Spleens were removed, and immune lymphocytes were fused with the rat myeloma cell line 210RCY3 Ag 1.2.3 (Y3). Supernatants were harvested and assayed for antibodies to p21 by GDP binding assay or immunofluorescence on Ha-SV-transformed MDCK cells, followed by immunoprecipitation testing.

Target Information

Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDK) and is transcriptionally activated by p53. It plays a key role in cellular response to genotoxic stress and is a major transcriptional target of p53.
In the nucleus, p21 binds and inhibits Cdk1 and Cdk2, blocking the G1 to S phase transition or G2 to mitosis after DNA damage, allowing DNA repair.
In the cytoplasm, p21 exhibits anti-apoptotic effects by inhibiting caspase 3, ASK1, and JNK.
P21 thus has dual roles in carcinogenesis—acting both as a tumor suppressor and as an oncogene.
