SIRT5는 Sirtuin(Sir2-like protein) 계열에 속하는 인간 단백질로, 원핵생물과 진핵생물 모두에서 발견됩니다. 이 단백질들은 보존된 sirtuin core domain을 가지며, NAD를 대사하거나 mono-ADP-ribosyltransferase로 작용할 수 있습니다. Sirtuin의 효소적 기능은 완전히 규명되지 않았으나, 최근 연구에서 Sir2에 의한 NAD 대사 및 히스톤 탈아세틸화 활성 간의 상호작용이 보고되었습니다. 이는 NAD와 아세틸화 히스톤의 N-epsilon-acetyl-lysine 잔기 간의 상호작용을 통한 활성 조절 메커니즘을 시사합니다.
