Target Information
Isoprenylation/methylation is an important posttranslational lipid modification involving the covalent addition of a 15-carbon farnesyl or a 20-carbon geranylgeranyl group to the C-terminus of a protein, resulting in increased hydrophobicity.
All known G proteins are modified in this way, making this pathway central to understanding signal transduction. From nematodes to humans, many eukaryotic proteins are reported to be isoprenylated.
Known prenylated proteins include:
- Fungal mating factors
- Nuclear lamins
- Ras and Ras-related GTP-binding proteins (G proteins)
- Subunits of trimeric G proteins
- Protein kinases
- At least one viral protein
Prenylation promotes membrane interactions of these proteins and plays a major role in protein-protein interactions.
