Cellular Localization Nucleus; Cytoplasm; Nucleus (PML body). Accumulates on chromatin in response to replication stress. Complexed with CCNT1 in nuclear speckles, but uncomplexed form in the cytoplasm. The translocation from nucleus to cytoplasm is XPO1/CRM1-dependent. Associates with PML body when acetylated.

Background

Cyclin dependent kinase 9 (CDK9) is a member of the cyclin-dependent protein kinase (CDK) family, which are key regulators of the cell cycle. CDK9 functions as part of the multiprotein complex TAK/P-TEFb, an elongation factor for RNA polymerase II-directed transcription. It phosphorylates the C-terminal domain of RNA polymerase II’s largest subunit. CDK9 forms a complex with cyclin T or cyclin K for regulation. The HIV-1 Tat protein interacts with CDK9 and cyclin T, suggesting a possible role in AIDS pathogenesis.
