Product Specific Information

This 4MEJJ monoclonal antibody recognizes mouse aryl hydrocarbon receptor (AHR).
AHR is a ligand-activated transcription factor mediating the toxic effects of environmental contaminants such as PCBs and TCDD. It also binds naturally occurring compounds from fruits, vegetables, and cellular metabolism.

AHR is localized in the cytoplasm in a complex with HSP90, p23, and XAP2/AIP/ARA9. Upon ligand binding, it translocates to the nucleus, forms a complex with ARNT, and binds to the DNA consensus sequence GCGTG, regulating genes such as CYP1A1.
Highest expression is observed in the liver, and AHR regulates differentiation of Treg and Th17 cells.

  • Recognizes b-1, b-2, and d alleles of mouse AHR; does not cross-react with human AHR.
  • Reported applications: Intracellular staining followed by flow cytometric analysis.
  • Tested with Th17-polarized mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
  • Recommended use: ≤0.5 µg/test (100 µL total volume).
  • Cell number: 10⁵–10⁸ cells/test (empirically determined).
  • Carefully titrate for optimal performance.

Target Information

AHR (Ah Receptor) belongs to a protein family including ARNT (HIF-1 Beta) and Drosophila PER and SIM proteins.
It contains an N-terminal PAS domain (~200 amino acids) and binds to DNA enhancer sequences to initiate CYP1A1 transcription.

AHR regulates xenobiotic-metabolizing enzymes such as cytochrome P450.
Ligands include aromatic hydrocarbons and halogenated compounds like TCDD.
In non-ligand-activated cells, AHR is complexed with HSP90 in the cytoplasm. Upon activation, it forms a heterodimer with ARNT, enabling DNA binding.

Diseases associated with AHR include eosinophilic fasciitis and seborrheic dermatitis.
