Product Specific Information

This CXNFT monoclonal antibody reacts with mouse NOS2 (inducible NOS, iNOS). Nitric oxide synthase enzymes catalyze the formation of nitric oxide from L-arginine through an NADPH- and oxygen-dependent mechanism. NOS2 is induced by bacterial endotoxins and inflammatory cytokines such as IFN-γ and TNF-α and is expressed in M1 macrophages and myeloid-derived suppressor cells.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested by intracellular staining of stimulated mouse peritoneal exudate cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00). Recommended ≤0.06 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Titrate for optimal performance.

Light Sensitivity: Protect from light; tandem dye sensitive to photo-induced oxidation.
Fixation: Samples may be stored up to 3 days at 4°C in the dark in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54).
Excitation: 488–561 nm
Emission: 775 nm
Laser Compatibility: Blue, Green, Yellow-Green
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

iNOS (Inducible Nitric Oxide Synthase) catalyzes the oxidation of L-arginine to L-citrulline and nitric oxide (NO). NO acts as a signaling molecule involved in vasorelaxation, neurotransmission, and immune responses. iNOS expression is induced by cytokines (IFN-γ, TNF, IL-1, IL-2) and LPS, leading to sustained NO production. It is found in macrophages, hepatocytes, synoviocytes, and smooth muscle cells. Dysregulation of iNOS is associated with diseases such as achalasia and impotence.
