Target Information

IFI16 (gamma-interferon-inducible protein 16) binds double-stranded DNA, preferentially to supercoiled and cruciform DNA structures. It is involved in transcriptional regulation and acts as a transcriptional repressor. IFI16 may regulate hematopoietic differentiation by activating unknown target genes and controls cellular proliferation through modulation of cell cycle regulatory factors such as p53/TP53 and retinoblastoma protein.
IFI16 also plays a role in innate immune response by recognizing viral dsDNA in the cytosol and nucleus. Upon binding viral DNA, it recruits TMEM173/STING and induces IFN-beta production. It is essential for activation of the IFR3 signaling cascade during human herpes simplex virus 1 infection, promoting heterochromatin assembly on viral DNA and inhibiting immediate-early gene expression and replication.
Additionally, IFI16 is involved in MTA1-mediated epigenetic regulation of ESR1 expression in breast cancer.
