In Western blots, no cross-reactivity with human p53 that is unphosphorylated at S15 is detected.
Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. p53 activation leads to the expression of p21^waf1 and other proteins involved in G1 or G2/M arrest.
The structure of p53 includes:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerisation domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is critical for p53 activation and stabilization. p53 plays a key role in preventing proliferation of damaged cells and acts as an anticancer agent. Alterations in p53, including mutations in the TP53 gene, are commonly associated with various human cancers and hereditary cancer syndromes such as Li-Fraumeni syndrome.
