Product Specific Information

Despite its predicted molecular weight, Syntaphilin often migrates at a higher molecular weight in SDS-PAGE.
A suggested positive control is rat brain tissue lysate.
PA5-20529 can be used with blocking peptide PEP-0649.


Target Information

Syntaphilin was initially identified in a yeast two-hybrid screen using the carboxy terminal region of Syntaxin-1 as bait.
Syntaxin-1 is a key component of the synaptic vesicle docking machinery forming the SNARE complex with synaptobrevin and SNAP-25.
Syntaphilin competes with SNAP-25 for binding to syntaxin-1 and inhibits SNARE complex formation, thereby regulating synaptic vesicle exocytosis.
It also binds dynamin-1 and inhibits dynamin-dependent endocytosis.
Mice lacking syntaphilin show increased mitochondrial motility and reduced axonal mitochondrial density, correlating with enhanced short-term facilitation and motor impairments, suggesting a major role in presynaptic function.
Multiple isoforms are known to exist.
