Target Information

Retinoic acid-inducible gene I (RIG-I) is a pattern recognition receptor (PRR) involved in detecting viral double-stranded RNA (dsRNA). Together with MDA5, RIG-I activates the innate immune response by recognizing viral RNA.

• RIG-I detects short dsRNA and dsRNA lacking a 5′-triphosphate end.
• MDA5 detects long dsRNA.
  Once activated, both signal through IPS-1, leading to activation of NF-κB and IRF-3 transcription factors, resulting in apoptosis, cytokine signaling, and inflammation.
  RIG-I plays a crucial role in antiviral responses against influenza A/B, RSV, paramyxoviruses, Japanese encephalitis virus, and West Nile virus.
  MicroRNA-146a negatively regulates RIG-I-dependent antiviral responses by targeting TRAF6, IRAK1, and IRAK2.
  Recent studies also suggest RIG-I detects cytosolic DNA via RNA polymerase III activity.
