The His tag is a short amino acid sequence consisting of histidine (His) residues appended to recombinant proteins. Ni-charged resins are used for purification of His-tagged proteins from eukaryotic and prokaryotic expression systems.

THE™ His Tag Antibody [iFluor 647], mAb, Mouse is THE™ His Tag Antibody (A00186) conjugated with iFluor 647 under optimal conditions with an F/P ratio of 3–6. It is suitable for detecting the expression level of His-tagged proteins. iFluor 647 is a bright and photostable fluorescent dye, serving as an excellent alternative to Alexa Fluor 647.

Target Information

The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH). It is commonly expressed at either the N- or C-terminal region of recombinant proteins to enable isolation or purification by immobilized metal affinity chromatography (IMAC).

Epitope tagging allows fusion of a known epitope to a recombinant protein for detection when no specific antibody is available. His-tagged proteins can be purified using metal chelate chromatography due to the strong affinity between histidine clusters and metal ions such as Ni²⁺.

His tag-specific antibodies are valuable tools for detection and co-immunoprecipitation of tagged proteins.
