Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA), expressed on all hematopoietic cells except erythrocytes and platelets. The cytoplasmic domain of CD45 functions as a tyrosine phosphatase critical for lymphocyte activation.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

• Applications Tested: Pre-diluted and validated for flow cytometry of human peripheral blood cells
• Recommended usage: 5 µL (0.8 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus per sample before combining with cells.
• For single-color controls: use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

NovaFluor conjugates are based on Phiton™ technology, utilizing novel nucleic acid dye structures engineered for optimized fluorescence performance with minimal spillover and spread.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, constituting about 10% of lymphocyte surface proteins. It is absent on non-hematopoietic cells and tissues. CD45 plays a key role in lymphocyte development, antigen receptor signaling, and regulation of Src-family kinases. Isoforms of CD45 arise from alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. CD45 is essential for T- and B-cell signaling and modulates integrin-mediated signal transduction, apoptosis, and cytokine receptor signaling.
