Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except circulating erythrocytes and platelets. It plays a key role in lymphocyte activation via tyrosine phosphatase activity.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells. Recommended usage: 5 µL (0.8 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining. Add 5 µL per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, featuring engineered nucleic acid dye structures designed to minimize spillover and spread effects.

Target Information

CD45 (LCA) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells. It regulates Src-family kinases and cytokine receptor signaling, crucial for lymphocyte development and antigen signaling. CD45 isoforms arise from alternative splicing and differ in extracellular domains but share identical transmembrane and cytoplasmic regions. It is a major regulator of T- and B-cell activation and survival.
