Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells excluding circulating erythrocytes and platelets. The cytoplasmic portion of CD45 has tyrosine phosphatase activity and plays a key role in lymphocyte activation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry of normal human peripheral blood cells at 5 µL (0.8 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor conjugates utilize Phiton™ technology, employing novel nucleic acid dye structures that minimize spectral spillover and spread.

Target Information

CD45 (LCA, Leukocyte Common Antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, comprising about 10% of lymphocyte surface proteins.
It is absent in non-hematopoietic cells and tissues. CD45 regulates T- and B-cell receptor signaling, cytokine receptor pathways, and integrin-mediated signaling, influencing immune cell activation, survival, and apoptosis.
CD45 isoforms arise from alternative splicing and differ in extracellular domains while sharing identical transmembrane and cytoplasmic regions.
