Product Specific Information

The CD28.2 monoclonal antibody reacts with the human CD28 molecule, a 44 kDa homodimer expressed by thymocytes, mature T cells, and plasma cells. CD28 binds CD80 (B7-1) and CD86 (B7-2) and serves as a potent co-stimulator of T cells. Signaling through CD28 enhances IL-2 and IL-2 receptor expression and increases cytotoxicity of CD3-activated T cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤1 µg/test (100 µL final volume, cell number empirically determined between 10⁵–10⁸ cells/test). Antibody should be titrated for optimal performance.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T cell surface glycoprotein belonging to the immunoglobulin superfamily. It is constitutively expressed on most peripheral blood T lymphocytes and acts as a critical costimulatory receptor providing the second activation signal via binding to CD80 and CD86 on antigen-presenting cells.

CD28 prevents T cells from entering an anergic state or undergoing premature apoptosis. In murine lymphoid organs, CD28 is expressed on all CD4⁺ and CD8⁺ T cells. Expression is highest on immature thymocytes and lower on mature cells.

Diseases associated with CD28 dysfunction include mycosis fungoides and Sezary’s disease.
