Target Information
The mitochondrial succinate dehydrogenase complex subunit D (SDHD) is one of four proteins that constitute the tricarboxylic acid (TCA) cycle enzyme succinate dehydrogenase (SCH).
Mutations in SDHD are associated with hereditary paragangliomas, which are typically benign tumors of the autonomic nervous system.
SDHD is suggested to act as a tumor-suppressor gene. In cases with nonsense mutations (e.g., R22X), loss of heterozygosity and complete loss of Complex II enzymatic activity have been observed, along with increased levels of angiogenic factors such as EPAS1 and VEGF, potentially promoting tumor growth.
