Target Information

MPP8 is a heterochromatin component that specifically recognizes and binds methylated Lys-9 of histone H3 (H3K9me), promoting recruitment of proteins mediating epigenetic repression. It mediates recruitment of the HUSH complex to H3K9me3 sites, maintaining transcriptional silencing by promoting SETDB1 recruitment and H3K9me3 deposition. MPP8 also recruits DNMT3A for DNA methylation at CpG sites, down-regulates CDH1 expression, and represses L1 retrotransposons in collaboration with MORC2 and SETDB1. The HUSH complex further silences unintegrated retroviral DNA via ZNF638 recruitment.
