STAT3 (Signal Transducer and Activator of Transcription 3)는 STAT 단백질 패밀리에 속하며, 수용체 관련 키나아제에 의해 인산화되어 핵으로 이동 후 전사인자로 작용합니다.
STAT3는 Interferon-alpha, Interferon-gamma, EGF, PDGF, IL6 등에 의해 활성화될 수 있으며, Tyr705에서의 인산화는 이합체 형성, 핵 이동, DNA 결합에 필수적입니다.
STAT3는 세포 성장, 세포사멸 조절 등 다양한 세포 과정에서 중요한 역할을 하며, Rac1 및 PIAS3 단백질과 상호작용하여 활성 조절을 받습니다.
인간의 STAT3 유전자는 염색체 17q에 위치하며, 세 가지 스플라이싱 변이체가 보고되어 있습니다.
Knock-out 마우스에서 STAT3 유전자 결실은 배아 단계에서 치명적임이 확인되었습니다.
