Target Information
Proteins of the Bcl-2 family are critical regulators of apoptosis. Proapoptotic members, such as Bax, contain three of the four Bcl-2 homology regions (BH1–3), while BH3-only proteins, such as Bim, possess only the short BH3 motif.
Database searches revealed Bfk, a novel member of the Bcl-2 family that contains BH2 and BH3 regions but lacks BH1 and BH4. Bfk is closely related to Bcl-G(L), lacks a C-terminal membrane anchor, and is cytosolic. Overexpression of Bfk weakly promotes apoptosis and antagonizes Bcl-2’s prosurvival function.
Low expression levels of Bfk are found in stomach, ovary, bone marrow, and spleen, but increase markedly in the mammary gland during pregnancy, suggesting a role in mammary development.
