Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by most thymocytes, a subset of mature T helper cells, and at low levels on monocytes. CD4 acts as a receptor for HIV; SK3 blocks HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Applications Reported:
This antibody is validated for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.06 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited by the blue laser (488 nm). Recommended filter: 710/50 bandpass (695/40 acceptable alternative). Ensure instrument compatibility.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD4 antigen recognizes MHC class II molecules and acts as a co-receptor for HIV. It is primarily expressed in T helper lymphocytes but also found in monocytes, macrophages, and dendritic cells. CD4 is detected in thymus, lymph nodes, tonsils, spleen, and various non-lymphoid tissues. It contributes to early T-cell activation via association with the T-cell receptor complex and Lck kinase, and may mediate neuronal damage in infectious or immune-mediated CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
