Product Specific Information

Description:
The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. CD3e, along with gamma and delta subunits, is essential for proper assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Binding of 145-2C11 to TCR initiates intracellular signaling pathways leading to activation, proliferation, or apoptosis depending on experimental conditions. Commonly used as a phenotypic marker for mouse T cells.

Used for flow cytometric analysis.

Applications Tested:
Validated by flow cytometric analysis of mouse splenocytes at ≤1.0 µg/test (100 µL final volume). Optimal titration recommended.

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance. Add 5 µL per sample for optimal results.

Target Information

The CD3 complex transduces antigen-recognition signals into T-cell cytoplasm and regulates TCR surface expression. CD3 subunits (gamma, delta, epsilon, zeta) are members of the immunoglobulin superfamily encoded on chromosome 11. They have cytoplasmic tails associated with signal transduction molecules via tyrosine-based motifs.
CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells.
Defects in CD3 genes are linked to CD3 immunodeficiency.
