Product Specific Information

The monoclonal antibody 1A8-Ly6g reacts with mouse Ly-6G (Gr-1), a 25 kDa GPI protein expressed exclusively by neutrophils. Ly-6G is involved in neutrophil infiltration, recruitment, and migration, and associates with B2 integrins CD11a and CD11b.

Unlike the RB6-8C5 antibody, 1A8-Ly6g binds specifically to Ly-6G on neutrophils and has been used for neutrophil depletion with reported antagonist activity.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse bone marrow cells at ≤1.0 µg/test (100 µL final volume). Optimal titration is recommended for each assay.

Fluorochrome: Brilliant Ultra Violet™ 737 (BUV737)

• Emission: 732 nm
• Excitation: 355 nm
• Laser: Ultraviolet (UV)

Ensure the cytometer is equipped for UV laser detection.

Staining Buffer Recommendation:
Use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer (Product # 00-4409-75) to reduce non-specific polymer interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in:

• IC Fixation Buffer (Product # 00-8222)
• 1-step Fix/Lyse Solution (Product # 00-5333)
  for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Methanol Compatibility:
Compatible with short-term methanol-based fixation (≤1 hour). Avoid long-term storage in methanol-containing buffers.
