Product Specific Information

This antibody detects endogenous levels of Progesterone Receptor only when phosphorylated at Serine 294.

Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. The PR mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with the establishment and maintenance of pregnancy.
PR exists as two forms: a 94 kDa protein (Form A) and a 120 kDa protein (Form B), resulting from alternative translation initiation sites. PR-B is transcriptionally active and responsible for activating genes for endometrial maintenance, pregnancy maintenance, and inhibition of ovulation.
PR-A lacks 165 amino acids at the N-terminus compared to PR-B, exposing a 140 amino acid inhibitory domain that represses steroid hormone transcriptional activity.
In its inactive state, PgR forms a multiprotein complex including heat shock proteins and immunophilins. Upon progesterone binding, conformational changes promote dimerization and binding to progesterone response elements (PRE), leading to transcriptional activation.
A null mutation in the PGR gene results in pleiotropic reproductive abnormalities.
