Product Specific Information

This SRBCZX monoclonal antibody recognizes signal transducer and activator of transcription 5 (STAT5) phosphorylated on tyrosine 694.
STAT proteins are activated by cytokine receptor binding, leading to phosphorylation by JAK family kinases. Following phosphorylation, STAT5 translocates to the nucleus to regulate gene transcription.
Phosphorylation on Tyr694 is essential for STAT5 dimerization, nuclear translocation, and DNA binding.

Specificity was confirmed by ELISA and flow cytometry.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested for intracellular staining of stimulated normal peripheral blood cells at 5 µL (0.25 µg) per test (100 µL final volume).
Cell number range: 10⁵–10⁸ cells/test (empirically determined).

Recommended Staining Protocol:
Use Protocol C (Two-step Fixation/Methanol).
Protocol A and B (cytoplasmic/nuclear staining) are not suitable.

Target Information

STAT5 alpha is a transcription factor activated by cytokines and growth factors. Upon phosphorylation, it forms dimers and translocates to the nucleus to activate gene transcription.
STAT5 alpha mediates responses to IL2, IL3, IL7, GM-CSF, erythropoietin, thrombopoietin, and various growth hormones.
Activation of STAT5 alpha is linked to tumorigenesis in myeloma and lymphoma associated with TEL/JAK2 fusion.
Mouse STAT5 alpha induces BCL2L1/BCL-X(L) expression, suggesting anti-apoptotic roles.
STAT5 alpha and beta share 96% sequence similarity and are co-expressed in most tissues, with differential expression in muscle and mammary tissue.
STAT5 alpha is involved in reproduction, lactation, immune function, and somatic growth.
