Target Information
GAPDH (Glyceraldehyde-3-phosphate dehydrogenase)는 해당과정(glycolysis)에 관여하는 촉매 효소로, 동일한 37 kDa 서브유닛 4개로 구성된 테트라머 형태로 존재합니다. NADPH 존재 하에서 1,3-bisphosphoglycerate를 glyceraldehyde 3-phosphate로 환원시키는 반응을 촉매합니다.
GAPDH는 대부분의 조직에서 높은 수준으로 발현되며, 해당과정 외에도 DNA 복제, 수리, 핵 RNA 수출, 막 융합, 미세소관 결합 등 다양한 세포 과정에 관여합니다. 또한 세포사멸(apoptosis), 노화 관련 신경퇴행성 질환, 암세포의 신경세포 사멸 등에서도 중요한 역할을 하는 것으로 알려져 있습니다.
GAPDH는 Alzheimer’s disease의 원인 단백질인 amyloid precursor protein 및 Huntington’s disease의 원인 단백질인 Huntingtin과 결합하는 것으로 보고되었으며, actin 및 tubulin과의 상호작용도 보고된 바 있습니다.
GAPDH는 대부분의 조직에서 높은 수준으로 발현되므로 Western blot 분석 시 단백질 로딩 컨트롤로 유용하게 사용됩니다.
