Target Information
CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein belonging to the scavenger receptor cysteine-rich superfamily. It functions as a receptor for the hemoglobin-haptoglobin complex, protecting tissues from oxidative damage and aiding in iron recycling through endocytosis and heme breakdown.
CD163 is expressed exclusively on human monocytes and macrophages, particularly in the late phase of inflammation. It is found on all circulating monocytes and most tissue macrophages, except those in the mantle zone and germinal centers of lymphoid follicles, interdigitating reticulum cells, and Langerhans cells.
Expression is higher on CD16⁺ monocytes and is upregulated by IL-10, IL-6, and dexamethasone. LPS and PMA induce shedding of CD163 from the cell surface. CD163 binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner, showing higher affinity for HP1F-type complexes. It also triggers intracellular signaling involving tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL-6 and CSF1.
