Target Information

UBXN1 is a ubiquitin-binding protein involved in the modulation of the innate immune response. It inhibits both the RIG-I-like receptors (RLR) and NF-kappa-B pathways. Upon viral infection, UBXN1 is induced and recruited to the RLR component MAVS, where it interferes with MAVS oligomerization and disrupts the MAVS/TRAF3/TRAF6 signalosome.
It also interferes with TNFα-triggered NF-kappa-B signaling by interacting with cellular inhibitors of apoptosis proteins (cIAPs), preventing their recruitment to TNFR1. Additionally, it associates with CUL1 to inhibit NF-kappa-B inhibitor alpha (NFKBIA) degradation, maintaining NF-kappa-B in its inactive state.
UBXN1 interacts with the BRCA1-BARD1 heterodimer, regulating its E3 ubiquitin-protein ligase activity by binding Lys-6–linked polyubiquitin chains. It is also a component of a complex coupling deglycosylation and proteasome-mediated degradation of misfolded proteins in the endoplasmic reticulum.
