Product Specific Information

The monoclonal antibody SolA15 recognizes mouse and rat Ki-67, a 300 kDa nuclear protein expressed during all active phases of the cell cycle (G1, S, G2, M) but absent in resting (G0) cells. Ki-67 redistributes from the nucleus to chromosomes during mitosis and serves as a marker for cell proliferation. In tumor studies, the Ki-67 labeling index indicates the proportion of proliferating cells and is used as a prognostic marker.

The SolA15 antibody also reacts with human, non-human primate, and canine Ki-67.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested on stimulated mouse splenocytes using the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00) following BestProtocols® Protocol B.
Recommended usage ≤ 0.25 µg/test in 100 µL final volume (10⁵–10⁸ cells/test). Titrate for optimal performance.

Fluorochrome Information:
Alexa Fluor® 700 emits at 723 nm, excitable with red laser (633 nm). Recommended optical setup: 685 LP mirror and 710/20 filter.
Excitation: 633–647 nm / Emission: 723 nm / Laser: Red.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases of the cell cycle. It associates with euchromatin and heterochromatin regions and shows cell cycle–dependent localization. Ki-67 is widely used as a proliferation marker since it is absent in G0 cells but increases through mitosis. It serves as a valuable marker for determining the growth fraction of neoplasms and correlates with tumor grade and prognosis.
