Product Specific Information
- PA1-1746 detects human, rat, mouse, bovine, canine, and porcine aggrecan neo G1.
- Successfully used in Western blot and immunofluorescence procedures.
- Detects a ~64 kDa protein representing the neoepitope sequence (NITEGE) generated by aggrecanase-mediated cleavage at Glu373-Ala374 of aggrecan core proteins.
- Immunogen: Synthetic peptide corresponding to residues C G G N(387) I T E G E(392) of human aggrecan.
Target Information
Aggrecan, also known as cartilage-specific proteoglycan core protein (CSPCP) or chondroitin sulfate proteoglycan 1, is encoded by the ACAN gene.
It is a key structural component of the extracellular matrix in cartilage, providing resistance to compression.
The human protein (~250 kDa, 2316 amino acids) exists in multiple isoforms due to alternative splicing and exhibits a bottlebrush structure with chondroitin sulfate and keratan sulfate chains.
Aggrecan degradation is associated with cartilage deterioration in joint injury, disease, and aging.
The protein includes three interglobular domains (G1, G2, G3) involved in aggregation, hyaluronan binding, cell adhesion, and chondrocyte apoptosis.
Aggrecan fragments are released into synovial fluid during osteoarthritis progression due to cleavage by aggrecanase and MMPs.
Antibodies BC-3 and BC-4 can detect neoepitopes generated by aggrecanase (ARGSVXXX) and MMP (XXXVDIPEN), respectively.
