Target Information
ROCK1은 세린/트레오닌 키나아제로, 세포 분화 및 이동을 조절합니다. Rho/ROCK 경로는 다양한 인체 암의 진행에 관여하며, ROCK1은 새로운 Caspase-3 기질로 밝혀졌습니다.
ROCK은 소형 GTPase Rho의 효과기로, myosin light chain (MLC)의 직접 인산화 및 MLC phosphatase의 인산화를 통해 세포 수축성을 증가시킵니다. 이는 세포사멸 과정 중 막 돌출(blebbing)을 유도하는 중요한 단계입니다.
ROCK1은 N-말단의 kinase domain과 C-말단의 억제성 cysteine/histidine-rich domain으로 구성되어 있으며, Rho-binding domain을 포함하는 가변 영역으로 연결되어 있습니다.
세포사멸 시 caspase-3에 의해 ROCK1의 억제 도메인이 절단되어 지속적이고 조절되지 않는 kinase 활성이 유도됩니다. 이는 MLC 인산화와 막 돌출을 촉진하여 세포사멸의 실행 단계에서 중요한 역할을 합니다.
