Target Information

This gene encodes a transmembrane receptor commonly referred to as DC-SIGN, expressed on dendritic cells and macrophages. It plays a key role in the innate immune system, recognizing diverse pathogens such as parasites and viruses. The protein contains three domains:

  • N-terminal transmembrane domain
  • Tandem-repeat neck domain
  • C-type lectin carbohydrate recognition domain

The extracellular region functions as both a pathogen recognition receptor and a cell adhesion receptor, binding carbohydrate ligands on microbial and host cell surfaces. The neck domain facilitates homo-oligomerization, enabling multivalent ligand binding with high avidity.
Rare variations in the number of 23 amino acid repeats in the neck domain significantly affect ligand binding.
DC-SIGN is closely related to L-SIGN (GeneID 10332) but differs in ligand-binding properties and tissue distribution.
Alternative splicing produces multiple variants.
