Target Information
Derlin-1 is a functional component of endoplasmic reticulum-associated degradation (ERAD) for misfolded lumenal proteins. It may form a channel that allows retrotranslocation of misfolded proteins into the cytosol for ubiquitination and degradation by the proteasome. Derlin-1 mediates interactions between VCP and degradation substrates. During cytomegalovirus infection, it facilitates export and degradation of MHC class I heavy chains via interaction with the US11 viral protein, and also participates in degradation of misfolded cytomegalovirus US2 protein.
