Target Information

Vaspin is a newly described adipocytokine expressed predominantly in visceral white adipose tissues. Structural analysis predicts the presence of three beta-sheets, nine alpha-helices, and one central loop, characteristic of Serpin family members. Serpins are irreversible serine-protease inhibitors sharing over 30% sequence homology with alpha1-antitrypsin and a conserved tertiary structure containing an exposed reactive center loop that acts as a pseudo-substrate for target proteinases.

Members of this family are involved in key biological processes such as blood coagulation, fibrinolysis, complement activation, angiogenesis, inflammation, and tumor suppression. In humans, serpins comprise approximately 2% of total serum proteins, with alpha1-antitrypsin accounting for 70%. Vaspin shows 40.2% sequence identity with alpha1-antitrypsin, though its protease inhibitory activity remains unknown.

Vaspin mRNA expression in visceral fat correlates positively with BMI and body fat percentage. Administration of vaspin to obese mice improved glucose tolerance and insulin sensitivity, normalizing blood glucose levels and reversing altered expression of diabetes-related adipocytokines such as leptin, adiponectin, resistin, and TNF-alpha. These findings suggest potential clinical applications for Vaspin in addressing metabolic syndrome-related abnormalities.
