Target Information

This gene encodes a transmembrane receptor commonly referred to as DC-SIGN, expressed on dendritic cells and macrophages. It functions in the innate immune system, recognizing a wide range of pathogens including parasites and viruses.
The protein consists of three main domains:

  • N-terminal transmembrane domain
  • Tandem-repeat neck domain
  • C-type lectin carbohydrate recognition domain

The extracellular region (neck and lectin domains) acts as both a pathogen recognition receptor and a cell adhesion receptor by binding carbohydrate ligands on microbial and host cell surfaces.
The neck domain enables homo-oligomerization, allowing high-avidity binding to multivalent ligands.
Variations in the 23 amino acid repeats within the neck domain can affect ligand binding.
DC-SIGN is closely related to L-SIGN (GeneID 10332), differing in ligand-binding properties and tissue distribution.
Alternative splicing results in multiple isoforms.
