Target Information
Tyrosine hydroxylase (TH)는 카테콜아민 신경전달물질인 도파민, 에피네프린, 노르에피네프린의 생합성에 관여하는 효소입니다.
모든 종에서 TH의 활성은 보조인자 tetrahydrobiopterin (BH4)과의 상호작용에 의해 조절되며, BH4는 TH의 촉매 도메인에 결합하여 효소 활성을 유도합니다.
인간에서는 네 가지 mRNA 스플라이스 변이체(hTH1–hTH4)가 존재하며, 모두 동일한 촉매 도메인을 가지지만 N-말단 조절 도메인에서 차이를 보입니다.
TH는 L-tyrosine을 L-dopa로 전환하는 역할을 하며, 아드레날린성 뉴런의 생리학적 기능에 핵심적인 역할을 합니다.
또한 TH의 기능 이상은 파킨슨병, 조현병, 세가와 증후군, 근긴장이상증 및 다양한 심혈관 질환과 관련이 있습니다.
