Product Specific Information
For reconstitution, add 100 µL distilled water to obtain a final antibody concentration of about 1 mg/mL.
For conjugation experiments, perform an additional desalting step using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).
Target Information
CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein and a member of the scavenger receptor cysteine-rich superfamily. It functions as a receptor for the hemoglobin-haptoglobin complex, protecting tissues from oxidative damage and aiding in iron recycling through endocytosis of hemoglobin/haptoglobin and heme degradation.
CD163 is expressed on human monocytes and macrophages, particularly in the late phase of inflammation. It is present on CD14⁺ and CD64⁺ monocytes, with higher expression on CD16⁺ monocytes. Expression is upregulated by IL-10, IL-6, and dexamethasone, while LPS and PMA induce shedding of CD163 into plasma or supernatant. CD163 binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner, showing higher affinity for HP1F-type complexes. It also triggers intracellular signaling cascades involving tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and IL6 and CSF1 secretion.
