Target Information
Thymidine kinase-2 (TK2) is a deoxyribonucleoside kinase that phosphorylates thymidine, deoxycytidine, and deoxyuridine, as well as antiviral and anticancer nucleoside analogs. Both recombinant and native forms exhibit broad substrate specificity and complex kinetics, suggesting a role in activation of chemotherapeutic nucleoside analogs.
Highest expression levels are found in testis and ovary. TK2 and deoxyguanosine kinase (DGK) provide the main supply of deoxyribonucleotides (dNTPs) for mitochondrial DNA synthesis through the salvage pathway. Mutations in these kinases are associated with mitochondrial DNA depletion, indicating their importance in maintaining balanced mitochondrial dNTP pools.
