Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human IFN-gamma. Anti-Human IFN-gamma-specific antibody was purified by affinity chromatography employing an immobilized Human IFN-gamma matrix.

Sandwich ELISA:
To detect Human IFN-gamma by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human IFN-gamma (500-P32Bt) as a detection antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant Human IFN-gamma.

Western Blot:
To detect Human IFN-gamma by Western blot, use at 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Human IFN-gamma is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

IFN gamma (Interferon gamma, Type II interferon)

• A macrophage activation factor and immune interferon produced primarily by T-lymphocytes and NK cells in response to antigens and cytokines.
• Dimeric glycoprotein (~45 kDa) composed of two 146 amino acid subunits.
• Appears as 25, 20, and 15.5 kDa bands on SDS-PAGE due to differential glycosylation.
• Species specific; human IFN gamma does not cross-react with mouse.
• Functions include antiviral and antiproliferative activity, induction of MHC I/II, macrophage activation, and enhanced immunoglobulin secretion.
• Acts via IFN gamma receptor I/II and JAK-STAT pathway.
• Expression is upregulated by IL-2, FGF basic, EGF, and downregulated by vitamin D3 or DMN.
• Mutations in IFN gamma gene are associated with aplastic anemia.
