Target Information
Guanylate cyclase-activating protein (GCAP1) is a Ca²⁺-binding protein that enhances cGMP synthesis in photoreceptors. Mammalian GCAPs share over 90% similarity, consisting of 201–205 amino acids with three conserved Ca²⁺-binding sites. The GUCA1A gene (GCAP1) is expressed solely in the retina. Mutations such as Tyr99→Cys and Pro50→Leu have been linked to cone dystrophy and variable retinal phenotypes. The mutant proteins retain cyclase activation ability but show reduced thermal stability and increased protease sensitivity, potentially contributing to retinal cell degeneration.
