The Human TSLP Simplex ProcartaPlex™ Kit measures thymic stromal lymphopoietin (TSLP) protein and is designed to be combinable with other Simplex kits, allowing creation of custom multiplex panels utilizing Luminex™ xMAP technology for protein detection and quantitation.
When combining multiple Simplex kits (i.e., not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is required per assay plate regardless of plex size.

ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on sandwich ELISA principles, using two highly specific antibodies binding to different epitopes of one protein to quantitate all protein targets simultaneously via a Luminex instrument.
These assays require as little as 25 μL of plasma or serum, or 50 μL of cell culture supernatant, with results available in about four hours.

ProcartaPlex assays utilize Luminex™ xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single sample from plasma, serum, cell culture supernatants, and other bodily fluids.

The Luminex beads are internally dyed with precise proportions of red and infrared fluorophores to create spectrally unique signatures identifiable by Luminex xMAP detection systems (e.g., Luminex 200, FLEXMAP 3D, MAGPIX).
Similar to a sandwich ELISA, matched antibody pairs identify the target protein. Each bead type is conjugated to antibodies specific for a single protein target, and detection occurs via biotinylated antibodies and streptavidin–R-phycoerythrin (RPE).

기술적 원리

Unlike ELISA, the capture antibody in ProcartaPlex assays is conjugated to a bead rather than adsorbed to a microplate well, allowing reagents to remain free-floating in solution.
Detection instruments like Luminex 200 use two lasers—one to identify bead spectral signatures and another to quantify RPE fluorescence, proportional to protein concentration.
