The Human LAG-3 Simplex ProcartaPlex Kit measures LAG-3 protein and is designed to be combinable with other Simplex kits, allowing users to create custom multiplex panels utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits, only one buffer kit (sold separately) is required per assay plate, regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on the sandwich ELISA principle, using two highly specific antibodies binding to different epitopes of one protein to quantitate multiple protein targets simultaneously via a Luminex instrument. These assays require as little as 25 μL of plasma or serum, or 50 μL of cell culture supernatant, and deliver analyzed results within four hours.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single sample. Each bead is internally dyed with precise proportions of red and infrared fluorophores to create unique spectral signatures identifiable by Luminex systems (e.g., Luminex 200, FLEXMAP 3D, MAGPIX).

Similar to sandwich ELISA, each bead is conjugated with antibodies specific to a single target protein. Bound proteins are detected using biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The amount of RPE fluorescence is proportional to the protein concentration.

Unlike ELISA, the capture antibody in the ProcartaPlex assay is conjugated to a bead rather than adsorbed to a microplate well, allowing reagents to remain in solution. The Luminex instrument uses dual lasers—one to identify bead type and another to quantify RPE fluorescence—enabling multiplexed profiling of more targets using less sample in the same time as a traditional ELISA.
