Product Specific Information
This STLALYV monoclonal antibody recognizes human survivin (Birc5), a member of the inhibitor of apoptosis (IAP) family. Survivin inhibits caspase activation, regulating apoptosis negatively, and plays a key role in mitosis and cytokinesis. It is highly expressed in tumors and fetal tissue but absent in terminally differentiated and non-proliferating cells.
Applications Reported:
Used for intracellular staining followed by flow cytometric analysis.
Applications Tested:
Pre-titrated and tested by intracellular staining of stimulated human peripheral blood cells using the Foxp3/Transcription Factor Staining Buffer Set. Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with a 488 nm blue laser. Recommended filter: 710/50 bandpass (695/40 acceptable alternative). Protect from light due to photo-induced oxidation sensitivity.
Fixation:
Samples can be stored in IC Fixation Buffer or 1-step Fix/Lyse Solution for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.
Filtration:
0.2 µm post-manufacturing filtered.
Target Information
Survivin (IAP4) is the smallest member of the IAP family, involved in cell signaling, division, and metabolism. It shows high expression in fetal development and tumors but low in adult tissues. Survivin associates with mitotic spindle microtubules during G2/M phase, regulating apoptosis and mitotic progression. Overexpression in cancer may lead to aberrant mitosis and resistance to apoptosis. It is also expressed in brain tissues following injury, suggesting roles in neural cell proliferation.
