Description:
The B1.23.2 monoclonal antibody recognizes human Human Leukocyte Antigens (HLA) B and C, which along with HLA-A correspond to MHC Class I proteins in humans. HLA-B and HLA-C are expressed on most cell types and form heterodimers with beta-2 microglobulin on the cell surface. These complexes are essential for presenting endoplasmic reticulum-derived peptides to immune cells. The antibody has been shown to inhibit binding of KIR2DL1 and LIR1.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use ≤0.5 µg per test (100 µL final volume). Cell number should be empirically determined (10⁵–10⁸ cells/test). Optimal performance requires titration for each assay.

Target Information

HLA-class I antigens are widely distributed on human nucleated cells. Expression levels may change under pathological conditions — for example, malignant cells may lose HLA-ABC, while hepatocytes in alcoholic hepatitis, biliary cirrhosis, and chronic active hepatitis may show enhanced expression.
HLA and MHC antibodies are critical tools in Immunopeptidomics, aiding in the identification and characterization of neoantigens using high-performance liquid chromatography coupled to tandem mass spectrometry.
