Product Specific Information
The monoclonal antibody HK1.4 recognizes mouse Ly-6C, a GPI-linked protein of the Ly6 family. Ly-6C is expressed on monocytes/macrophages, endothelial cells, granulocytes, and subsets of lymphocytes. Expression varies among mouse strains:
- Ly6c.1 (C57Bl/6, SJL): stains both CD4 and CD8 cells
- Ly6c.2 (BALB/c, 3H/He): stains CD8 but not CD4 cells
In vitro, HK1.4 antibody can enhance proliferation and stimulate cytokine release.
Applications Reported: Flow cytometric analysis and functional assays.
Applications Tested: Flow cytometric analysis of mouse splenocytes, ≤0.25 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Antibody should be titrated for optimal performance.
Storage and Handling: Use in a sterile environment.
Filtration: 0.2 µm post-manufacturing filtered.
Purity: >90% (SDS-PAGE).
Endotoxin Level: <0.001 ng/µg antibody (LAL assay).
Aggregation: <10% (HPLC).

Granulocytes are white blood cells containing cytoplasmic granules, also known as polymorphonuclear leukocytes (PMN/PML). Their nuclei are typically lobed into three segments. Neutrophil granulocytes are the most abundant type. Granulocytes are released from bone marrow under regulation by complement proteins.
