Target Information
TET2 is a dioxygenase that catalyzes the conversion of 5-methylcytosine (5mC) to 5-hydroxymethylcytosine (5hmC), playing a key role in active DNA demethylation. It mediates subsequent conversion of 5hmC into 5-formylcytosine (5fC) and 5-carboxylcytosine (5caC), constituting the first steps in cytosine demethylation.
TET2 also recruits O-GlcNAc transferase (OGT) to CpG-rich transcription start sites, promoting histone H2B GlcNAcylation. Mutations in TET2 are associated with Polycythemia vera and Myelodysplastic syndrome.
