Product Specific Information

The SIDI8BEE monoclonal antibody reacts with human and rhesus macaque CD8 beta. The CD8b chain forms a heterodimer with the CD8a chain and is expressed on thymocytes and peripheral cytotoxic αβ TCR T cells. The CD8 receptor binds to MHC class I and is involved in T-cell development and activation.
SIDI8BEE detects CD8β in both CD8αβ heterodimers and CD8ββ homodimers and cross-reacts with human and rhesus macaque CD8b.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry of human peripheral blood lymphocytes.
Use 5 µL (0.8 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01)

• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample to antibody cocktails before combining with cells
• For single-color controls, add 5 µL per 100 µL cell sample (10³–10⁸ cells)

Target Information

CD8B is a cell surface glycoprotein expressed on cytotoxic T lymphocytes. It acts as a coreceptor with the T-cell receptor to recognize antigens presented by class I MHC molecules on antigen-presenting cells.
The functional CD8 receptor can form αα homodimers or αβ heterodimers, both structurally similar to immunoglobulin light chains.
The CD8B gene encodes beta chain isoforms, with multiple splice variants and a pseudogene on chromosome 2.
